We had developed ultra-small nanoprobe for measuring extracellular oxygen consumption rate (OCR). The reagent has excellent biocompatibility and stability in water. It can be stored in a solution at 4 ℃ for more than 1 year without obvious sedimentation or aggregation. The product has been sterilized before delivery and can be used directly without any treatment. Extracellular OCR reagents have excellent oxygen response sensitivity (as shown in the middle figure), and can achieve continuous measurement of oxygen concentration outside cells or in culture media via fluorescence intensity and time-gated fluorescence lifetime measurement techniques. They are an ideal tool for extracellular OCR measurement. As shown in the right figure, extracellular OCR reagents can be customized with different wavelengths and fluorescence lifetimes, we also offer customized solution according to your special needs in order to avoid spectral overlap with the other cell-staining dyes.

The extracellular oxygen consumption rate (OCR) reagent has been proven not to be endocytosed by living cells and can be stably suspended in the culture medium for long duration. By continuously measuring the oxygen consumption rate outside the cell, the mitochondrial function state of live cells can be obtained, as shown in the left figure. By regulating the mitochondrial function of live cells in the culture medium by adding mitochondrial inhibitors, the oxygen consumption rate (OCR) of cells can be effectively regulated, thereby achieving non-invasive and non-disruptive cell OCR analysis without affecting the normal energy metabolism of cells. The reagent has a fast response, high accuracy, and high measurement throughput, and is a powerful tool for live cell mitochondrial function assessment and energy metabolism analysis. With the help of Electroporation technology, the reagent can quickly diffuse into the cell fluid and realize real-time measurement of oxygen concentration in the cytoplasm.
細(xì)胞外OCR試劑不僅適用于高通量的96孔板進(jìn)行細(xì)胞、藥物和培養(yǎng)條件的篩選,也能通過(guò)電穿孔Electroporation技術(shù)導(dǎo)入到細(xì)胞液中,進(jìn)行細(xì)胞液中的顯微成像,實(shí)現(xiàn)單/活細(xì)胞細(xì)胞液中氧濃度的測(cè)量,對(duì)類器官、組織切片也有很好的適用性。這種試劑還可以被噴灑到植物葉片上,進(jìn)行植物光合作用的分析和研究。
The extracellular OCR reagent is not only suitable for high-throughput 96-well plate screening of cells, drugs, and culture conditions, but can also be useful in studying oxygen concentration in cytoplasm via electroporation. The reagent enables the measurement of oxygen concentration in single/live cells in the extracellular space, in cytoplasm, and is also suitable for organoids, tissue sections. This reagent can also be sprayed onto plant leaves for analysis and research on plant photosynthesis.
